Objevy Alexandra Fleminga
Život Alexandra Fleminga byl plný neuvěřitelných okolností, náhod, a objevů, díky kterým se stal posléze světoznámým, uznávaným mikrobiologem a bakteriologem. Jeho úspěšná vědecká kariéra započala už před První světovou válkou, ve které se vyznamenal na poli chirurgie, pokračovala v meziválečném období, ve kterém se uskutečnily jeho nejvýznamnější pokusy a byla zakončena ziskem Nobelovy ceny a pasováním do šlechtického stavu Sir.
Objevy v mládí
Jak už bylo zmíněno v úvodu, život Fleminga byl samá náhoda. Po ukončení lékařské fakulty byl vystudovaný chirurg, ale díky profesorům Wrightovi a Freemanovi, který si ho vybral pro jeho talent, ne v bakteriologii, ale ve střelbě, se stal mladý Alexandr součástí laboratoře, ve které působil celý život. První větší úspěch bylo stanovení prvotních příznaků tuberkulózy, čímž si laboratoř zajistila jméno a příjmy. Toto velice slibné období ukončila Světová válka.
První světová válka
Flemingovo jméno se během Světové války zviditelnilo opět díky profesoru Wrightovi. Wright byl nasazen v laboratoři, která působila na území Francie. S sebou si povolal i Freemana s Flemingem. Zde se projevil Fleming jako chirurg, když začal praktikovat odebírání poškozené tkáně, aby se zabránilo infekci nebo jejímu případnému šíření. Profesor Wright zde dokázal, že ke zničení mikrobů je třeba zavést do rány hypertonický roztok soli, který zvyšuje odtok lymfy z těla. Dalším významným počinem této laboratoře bylo zjištění, že prostředky, kterými se sterilizovali chirurgické nástroje, jsou nedostačují, ba dokonce napomáhají růstu a životu bakterií. Objev lysozymu
K tomuto objevu došlo v roce 1922. Fleming vzal nosní hlen a umístil ho do Petriho misky, ve které byli už předtím kolonie mikroorganismů, které následně uhynuly. To samé se stalo, když umístil do misky slzu. Proto se rozhodl zkoumat slzy a nalezl enzym, který pojmenoval lysozym. Během dalších pozorování zjistil, že lysozym nezabíjí všechny mikroorganismy, ale pouze ty neškodné. Ovšem Fleminga napadlo, že lysozym vlastně určuje, které mikroorganismy jsou patogenní. Bakterie povětšinou zabíjel lysozym obsažený ve vaječném bílku, který ovšem nikdo neuměl oddělit. Proto pokus probíhal aplikací ředěného bílku nitrožilně králíkovi. Pokusy byly úspěšné, ale příliš náročné na tehdejší dobu.
Lysozym (lysis-zředěný) je enzym, který se vyskytuje v lidském těle nebo vaječném bílku. Má silné antibakteriální účinky, které narušují buněčné stěny bakterií.
Objev penicilinu
Je známo, že největší objevy a vynálezy vznikly a vznikají náhodou. Ne jinak tomu bylo v roce 1928 kdy, se přepsala historie medicíny, aniž by to kdokoliv v té době věděl nebo snad jenom tušil. Alexander Fleming toho dne vešel do laboratoře, kde byla zapomenutá Petriho miska, ve které byl agar (živná směs z červených řas, ve které se pěstují mikroorganismy), na něm byla plíseň. A právě v tento okamžik objevil Fleming antibiotickou účinnost penicilinu, konkrétně to byla houba Penicillium notatu. Látky, které uvolňovala, zabíjely většinu bakterií, kromě těch, které způsobují tyfus. Tento jev byl sice už známý, ale nikomu se ho nepodařilo provést laboratorně s takovým úspěchem. Aby mohli i nadále pozorovat penicilin, pěstoval se v masovém vývaru o teplotě 37⁰C. Po poradě s mykology byla tato teplota snížena na 20⁰C. Vzniklý vývar opravdu fungoval, zabíjel škodlivé bakterie a neškodil fagocytům (buňka, která je schopná pohlcovat pevné částice skrz membránu). Tento produkt fungoval při zředění, ale vydržel jen krátkou dobu. Zásadní problémem bylo že, nikdo neuměl oddělit tuto látku a vývar jako takový byl v medicíně nepoužitelný. Ačkoliv Fleming svůj objev publikoval mezi ostatní vědce, nikdo nejevil o jeho objev zájem.
Vše se změnilo v roce 1940 chemikovi Ernstu Chainovi a biochemikovi Howardu Floreymu. Brzy se začalo s testováním na zvířatech, které bylo více než uspokojivé. V únoru 1941 byl poprvé podán lék člověku, který umíral. Pro nedostatek látky pacient zemřel, ale léčivá účinnost penicilinu byla prokázána i na lidech. Po dostatečném nashromáždění zásob, byl penicilin podán dalším třem lidem. Dva pacienti se úspěšně uzdravili, třetí zemřel na komplikace.
Naneštěstí pro Fleminga i společnost se tyto převratné objevy odehrály během Druhé světové války. Nikdo si nemohl dovolit zafinancovat výrobu léčiva, která by byla prodělečná. Proto se Florey rozhodl odjet do USA, kde se pokusili vypěstovat penicilin na kukuřici, ale to se nezdařilo. Jediným úspěchem byl plesnivý meloun, který se stal základem na výrobu penicilinu v USA a prvních deseti tisíců litrů. Z této kultury vznikly i všechny ostatních, na kterých se vyrábí penicilin dodnes. V roce 1942 se rozšířila zpráva o novém léku a o rok později už se o něj zajímali farmaceutické společnosti. Tento objev vynesl pánům Chainovi, Floreymu a Flemingovi Nobelu cenu, kterou obdrželi 25.10.1945.
Sir Alexander Fleming byl za svůj bohatý život ověnčen mnoha oceněními a zásluhami. On sám však sebe považoval vždy za člověka náhod. A jak už bylo mnohokrát řečeno, tohoto velikána medicíny doprovázela náhoda na každém kroku. A za to jsme vděčni.
